Genetics shape who survives infections once thought harmless

A Nature News feature explains that inherited immune-system mutations—and, more recently, autoantibodies against interferons—help determine why some people die from microbes that are usually harmless. Researchers have cataloged hundreds of inborn errors of immunity (IEIs) across many genes, with defects like interferon-γ receptor mutations causing severe mycobacterial disease, and autoantibodies linked to severe COVID-19 and other infections. Some mutations can even confer protection against specific pathogens (e.g., CCR5 against HIV). With incomplete penetrance and epigenetic regulation, IEIs can vary by cell type and over a person’s lifetime, prompting genetic screening and targeted therapies to prevent or treat severe infections.

Gene therapy startup aims to use viruses to fight cancer.

A new startup founded by a former University of California San-Francisco investigator, Nicole Paulk, is using docile viruses developed for delivering healthy genes to patients with genetic diseases to slide interferons and other immune-stimulating molecules into unsuspecting cancer cells. The startup aims to turn the workhorse of gene therapy against cancer by using interferon, a protein from the body's immune system, to stimulate the immune system to fight cancer.

Immune system mechanism fights parasitic infection.

Penn State immunology researchers have discovered that type I interferon, a protein that is important for the body's defense against viruses, plays a critical role in suppressing inflammation in mice infected with the schistosome parasite. Schistosomiasis is a parasitic disease responsible for more than 250 million human infections worldwide. The discovery may lead to effective therapies for those suffering from schistosomiasis, second only to malaria as the most prevalent parasitic disease globally.
