A multi-dataset analysis links the APOE gene, especially the ε3 and ε4 variants, to 72–93% of Alzheimer's cases and about 45% of dementia cases, suggesting APOE and its pathways as major targets for prevention and treatment while noting that genetic risk interacts with lifestyle and environmental factors and is not determinative.
