"Intramolecular Bivalent Glues for Targeted Protein Degradation"
A recent study describes a new compound, IBG1, which effectively degrades BRD4, a protein associated with cancer, independently of the previously targeted protein DCAF15. The compound recruits the CRL4-DCAF16 complex to degrade BRD4, and it enhances the affinity of DCAF16 for BRD4. The study also reveals that IBG1 bivalently binds both bromodomains of BRD4, and it selectively degrades BRD2 and BRD4 but not BRD3. These findings provide insights into a potential new approach for targeted protein degradation in cancer therapy.
